摘要
【目的】研究双氢青蒿素对人红白血病K562细胞DNA的损伤作用。【方法】体外培养人红白血病K562细胞,加不同浓度双氢青蒿素处理24 h后,MTT法测定IC50;用单细胞凝胶电泳技术观察双氢青蒿素对K562细胞DNA的损伤情况。【结果】双氢青蒿素处理细胞后,MTT检测IC50为8×10-5mol/L;单细胞凝胶电泳显示双氢青蒿素作用24 h后可见明显的彗星状拖尾,而对照组细胞呈现明亮圆点。【结论】双氢青蒿素可抑制K562细胞的生长,对其DNA有致损效应。
[Objective] To investigate the effect of damage of dihydroartemisinin (DHA) on leukemic K562 cell DNA. [Methods] The DNA damage of K562 ceil induced by DHA was studied in vitro by MTT and single ceil gel electrophoresis. [Results] K562 ceils were treated with DHA of different concentrations, the IC50 value was 8 ×10^-5 mol/L. Single ceil gel electrophoresis showed comet-like tails, but control group showed bright dot. [Conclusions] DHA can inhibit the proliferation and damage of DNA of K562 ceils.
