摘要
建立了喜树果中喜树碱的高效毛细管电泳安培检测方法。着重探讨了缓冲溶液种类、浓度、酸碱度及操作电压、进样时间等对检测的影响。Na2B4O7-NaOH(Na2B4O7浓度为30mmol/L)为电泳介质,在15kV高压,pH9.0的碱性条件下片柱端安培检测喜树碱含量。该法的线性范围为:1-100mg/L,Y=11020+1014.6P,r=0.9994,检出限为0.5mg/L。
Camptothecin from Fructus Camptothecae Acuminatae was determined by capillary electro- phoresis with amperometric deteetion(CE/AD). The buffer pH value, the types of buffer solution, the concentration of the buffer, the applied voltage and the sampling time were selected as the investi-gation variables. Under the optimized eonditions: a running buffer composed of 30 mmol/L Na2B4O7 at pH 9.0, separation voltage at 15 kV, injection time for 10 s and detection voltage at 0.8 V, a line-arity exists between peak area(Y) and mass eoncentration of camptotheein(p) in the range of 1-100mg/L, Y= 11020 + 1014.6 p, r =0.9994. The deteetion limit is 0.5 mg/L.
