摘要
目的建立测定马钱子碱脂质体包封率的方法。方法采用紫外分光光度法测定马钱子碱的浓度,凝胶柱层析法分离马钱子碱脂质体和游离的马钱子碱。结果紫外分光光度法测定脂质体中马钱子碱浓度的回收率为100.61%~101.75%,日内精密度≤0.59%,日间精密度≤1、99%。凝胶柱层析法可以实现基线分离,马钱子碱脂质体上柱后的回收率为95.54%~99.84%。结论本方法准确度高,可以满足马钱子碱脂质体质量控制的需要。
OBJECTIVE To establish the method of determining the packaging ratio of brucine liposome. METHOD Ultraviolet spectrophotometry was used to determine the concentration of brucine and gel column stratographic analysis was used to isolate brucine liposome and the free brucine. RESULT The recovery rate of brucine concentration was found to be 100.61% ~ 101.75 %, with precision in a day≤0.59% and daytime precision≤ 1.99%. Gel column stratographie analysis could be used for basic line isolation, and the recovery rate of brueine liposome was 95.54% ~ 99.84%. CONCLUSION This method is highly accurate and can be used in the quality control of brueine liposome.
