摘要 目的建立用RP-HPLC法测定生物体内士的宁浓度的方法。方法色谱柱为DIKMA Diamonsil^TM C18(250mm×4.6mm,5μm),流动相为乙腈-0.01%三乙胺溶液(用磷酸调节pH值至2.6)(12,88),流速为1.0mL/min,检测波长为254nm。结果士的宁在0.75~18.0μg/mL的浓度范围内与峰面积呈良好的线性关系,在血液和肝脏中的回归方程分别为:Y=106751X-17366,r=0.9996和Y=118967X-25910,r=0.9996(n=7)。3种不同浓度的日内、日间RSD均小于10.0%。回收率在90%~110%,血液及肝脏中士的宁的定量下限分别为0.15μg/mL和0.45μg/g。绪论此方法准确、灵敏,能够用于测定生物体内士的宁的浓度。 Objective To establish a RP-HPLC method to determine the concentration of strychnine in biological materials. Methods DIKMA DiamonsilTM C18 column (250 mm× 4.6 mm, 5 μm) was used as an analytical column. The mobile phase consisted of 12 volume of acetonitrile, 88 volume of 0.01% triethylamine (adjusted to pH 2.6 with phosphoric acid). The flow rate was 1.0 mL/min. The UV detection wavelength was set at 254 nm. Results Strychnine had a good linearity in the range of 0.75-1.8μg/mL. The minimum detection concentration in blood plasma and liver was 0.15 μg/mL and 0.45μg/g, respectively. The recoveries of three different concentrations ranged from 90% to 110%, and the RSD values of Intra-day and inter-day were all under 10.0%. Conclusion The method is quick, precise and simple. It can be used to determine the concentration of strychnine in biological materials.
