摘要
蚕豆是高蛋白含酚类物质较多的作物之一,为了提取适于蚕豆AFLP分析的高质量的DNA,采用改良CTAB法,以β-巯基乙醇和PVP浓度为因素进行了实验。结果表明,用改良的CTAT法提取蚕豆DNA的质量较好,纯度高λ=260nm/280nm的平均OD值为1.915,变幅在1.81～2.08之间,绝大多数集中在1.9左右,其中,在CTAB提取液中加入1%的β-巯基乙醇和0.5%的PVP或直接加入1%PVP后,提取的DNA纯度较高,λ=260nm/280nmOD平均值分别为1.868和1.865,提取的DNA适于AFLP分析。
Faba bean is one of the high-protein crop containing phenol, in order to extract the high quality DNA from faba bean suitable for AFLP analysis, the modified CTAB procedures with the different concentions of β-mercaptoethanol and PVP was used in this study, the results showed that the DNA extracted by modified method was high quality and purity from faba bean ,the average OD value is about 1.915, ranging from 1.81 to 2.08, the majority of them were concentrated around 1.9, High quality DNA would be extracted with 1% β-mercaptoethanol and 0.5% PVP or only 1% PVP in CTAB, which the average OD valuve could be 1.868 and 1.865 respectively under the λ=260 nm/280nm, The extracted DNA could be used to do AFLP analysis.
