摘要
目的研究蜈蚣提取物对宫颈癌HeLa细胞生长的抑制及其作用机制。方法观察不同浓度的不同蜈蚣提取物对宫颈癌HeLa细胞的生长抑制情况:运用体外细胞培养技术,以不同浓度的蜈蚣提取物以及阳性对照药顺铂作用于培养的HeLa细胞48h,采用MTT法测定细胞代谢率,以流式细胞术观察DNA含量和凋亡的变化情况,DNA片断化分析和western Blot测定Bax,Caspase 3蛋白水平的表达。结果HeLa细胞用4,8,16mg/ml的乙醚、乙醇蜈蚣提取物处理48h后,细胞存活率显著降低;DNA周期改变明显,亚G1峰与对照组有显著性差异;DNA-Ladder典型;Western Blot提示蜈蚣提取物作用HeLa细胞后Bax,caspase 3蛋白表达,凋亡率显著增高。结论中药蜈蚣乙醚、乙醇提取物在体外对宫颈癌HeLa细胞的生长有明显地抑制作用,机制与改变细胞DNA周期,促进凋亡有关。
Objective To study the inhibitory effects and mechanism of centipede extract on proliferation of cervical cancer cell line HeLa. Methods To observe irthibitory - effect of different concentration extract on the growth of HeLa cell. By the method MTF to detect cell survival rate, by flow cytometry to observe the change of DNA cycle and apoptosis and to assay the DNA fragmentation and detect the protein Bax, caspase 3 by Western Blot in vitro cell culture. Results After HeLa cell had been treated with concentration centipede extract by aether or ethanol (4,8 , 16 mg/ml)for 48h, the survival rate decreased. The number of each cycle cells changed obviously, the sub -G1 peak between test and control group was significantly different, DNA -Ladder was typical, the protein Bax, Caspase 3 expressing, apoptosis rate increased significantly. Conclusion Centipede extract can in- hibit the growth of HeLa cell which is related to inhibiting DNA synthesis, blocking cancer cell division growth and enhancing apoptosis in vitro.
