摘要
目的探讨中药瑞香狼毒水提取物对肺癌细胞超微结构的影响。方法用瑞香狼毒水提取物处理两个肺癌细胞株(人小细胞肺癌NCI-H446和人非小细胞肺腺癌NC I-H157)。用药前后分别取材,按悬浮细胞操作规程常规制备超薄切片,透射电镜观察用药前后两肺癌细胞株超微结构的变化。结果用药后较用药前两肺癌细胞株的超微结构变化主要表现在:①细胞形态的改变:多数细胞体积变小,形态不规则,胞质电子密度增高。少数细胞体积增大,表面胞膜突起消失,部分细胞膜呈线性中断。②细胞器的改变:细胞器减少或变形、结构不清。个别细胞线粒体呈气球样肿胀,嵴减少或消失。③细胞核的改变:细胞核内染色质边集,基质电子密度增高。有些细胞核内异染色质分界不清,核仁呈均一电子密度,网状结构消失。结论中药瑞香狼毒水提取物对肺癌细胞有明显的细胞毒作用。
Objective To study the effects of aqueous extract from Ruixianglangdu on ultrastructure of lung carcinoma, Methods Two lung carcinoma cell strains were treated with aqueous extract from Ruixiangiangdu, the preparation of electric microscope （EM） samples were accomplished before and after being treated respectively. To compare the changes in ultrastructure of lung carcinoma between before and after being treated with aqueous extract from Ruixianglangdu. Results The changes of ultrastructure in lung carcinoma cell strains after being treated with extracts from Ruixianglangdu were shown：Firstly, in majority the size of lung carcinoma cells were contractible, and with irregular cellular morphous, the electron density of cytoplasm increased. In few of the lung carcinoma cells were swelling, the protuberance on cellular surface disappeared, in few of its, the cellular membrane were broken. Secondly, the cellular organdie of lung carcinoma cells decreased or deformation and unclear in structure. The mitochondria balloon swell, the mitochondrial cristae decreased or disappeared. Thirdly ,The nuclear chromatin of lung carcinoma cells concentrate to periphery, the electron density of cytoplasm increased, in some cells,the heterochromatin of nucleus were blurry,and the nucleole became uniform in electron density, the formatio reticularis of nucleole disappeared. Conclusion The extracts of Ruixianglangdu have obvious cytotoxicity to lung carcinoma cell strains.
