摘要
目的:分析非小细胞肺癌细胞株中人类表皮生长因子受体-2(c-erbB-2)基因的mRNA水平和蛋白水平。方法:采用RT-PCR测定肺腺癌(CALU-3和A549)和鳞癌NCL-H520细胞株c-erbB-2 mRNA水平;采用流式细胞术检测CALU-3、A549和NCL-H520细胞株c-erbB-2蛋白水平。结果:肺腺癌CALU-3细胞株c-erbB-2mRNA水平均高于腺癌A549和鳞癌NCL-H520细胞株,差异有统计学意义(P<0.05),而A549和NCL-H520细胞株之间差异无统计学意义(P>0.05);CALU-3细胞株c-erbB-2蛋白的阳性率均高于A549和NCL-H520细胞株,差异有统计学意义(P<0.05),而A549和NCL-H520细胞株之间差异无统计学意义(P>0.05)。结论:c-erbB-2基因在肺腺癌CALU-3细胞株中的表达均高于腺癌A549和鳞癌NCL-H520细胞株。
Objective：To investigate the expression of epidermal growth factor receptor-2 （c-erbB-2） mRNA and protein in different non-small cell lung cancer cell lines. Methods：C-erbB-2 mRNA and protein expression of the human lung adenocarcinoma cell lines （CALU-3 and A549） and squamous cell carcinoma NCL-H520 cell line with RT-PCR and flow cytometry were measured respetively. Results：The value of c-erbB-2 mRNA relative expression in CALU-3 cell line was 0.77±0.07 （F=5.488,P〈0.05）,significantly higher than in the cell lines A549 （0.65±0.05,P〈0.05） and NCL-H520（0.62±0.05,P〈0.05）. There was statistical signifcance in the difference between A549 and NCL-H520 cell lines （P〉0.05）. The c-erbB-2 protein positive expression rate was 33.78%±13.45% in the cell line CALU-3,significantly higher than in the cell lines A549 （15.60%±6.37%, P〈0.05） and NCL-H520 （4.22%±0.10%, P〈0.05）.There was no significant differences between A549 and NCL-H520 cell lines （P〉0.05）. Conclusion：Expression of c-erbB-2 gene in CALU-3 cell line was higher than in A549 and NCL-H520 cell lines.
