摘要
目的:从羊膜和羊膜细胞外基质结构特征、两者在组织工程中作为细胞支撑物负载细胞在实验研究、临床应用及羊膜上皮细胞可能是组织工程技术一种新的细胞来源等方面对该领域的研究现状进行综述,并分析了目前存在的问题及未来应用前景。资料来源:应用计算机检索Medline 2000-01/2007-03相关羊膜与羊膜细胞外基质在组织工程中应用方面的文献,检索词"amniotic membrane,tissue engineering",限定文献语言种类为English;同时检索中国期刊全文数据库、万方数据库2000-01/2007-03相关文献,检索词"羊膜,组织工程",限定文章语言种类为中文。资料选择:对资料进行初审,选取符合研究要求的有关文章找全文。纳入标准:①有关羊膜与羊膜细胞外基质结构特征的研究。②羊膜与羊膜细胞外基质作为组织工程支架材料的研究性论文。③有关羊膜细胞作为组织工程种子细胞的研究。排除标准:相关度不大的文章,重复性的文章。资料提炼:共检索到90篇关于羊膜与羊膜细胞外基质在组织工程中应用的文献,最终纳入30篇符合标准的文献。资料综合:①羊膜与羊膜细胞外基质结构特征:正常人羊膜组织结构分为上皮细胞层、基底膜、致密层、纤维细胞层及海绵层。上皮细胞层含有羊膜上皮细胞和由羊膜上皮细胞分泌的多种生长因子。羊膜细胞外基质:新鲜制备的羊膜细胞外基质与中、长期保存羊膜细胞外基质致密层结构均相似。②羊膜与羊膜细胞外基质作为组织工程支架材料的应用:羊膜与羊膜细胞外基质可以作为构建周围神经组织工程、皮肤组织工程、角膜组织工程、肌腱组织工程及骨组织工程的修复材料。③羊膜细胞作为组织工程种子细胞的应用:羊膜细胞成分有羊膜上皮细胞,成纤维细胞及未分化的间充质细胞。④分析了羊膜与羊膜细胞外基质在组织工程中应用目前存在的问题及未来应用前景。结论:人羊膜与羊膜细胞外基质作为一种生物材料,其取材、保存和使用需遵循严格的医学标准,但目前尚无统一检测标准。随着生物学技术和其他相关技术的发展,羊膜与羊膜细胞外基质在组织工程中的应用潜能必将得到不断的展现。
OBJECTIVE: To summarize the study status on the application of amniotic membrane and amniotic extracellular matrix in tissue engineering in recent years from their structure, their application in experimental study and clinic as cell support carriers, and amniotic endothelial cells as one novel call source.
DATA SOURCES: A computer-based online search of Medline was undertaken to identify the articles about the application of amniotic membrane and amniotic extracellular matrix in tissue engineering dated from January 2000 to March 2007 by using the keywords of "amniotic membrane, tissue engineering". Meanwhile, we searched China Periodicals Full-text Database and Wanfang database for the related Chinese articles published between January 2000 and March 2007 with the same keywords in Chinese.
STUDY SELECTION: Only those articles that ①contained information about the structure of amniotic membrane and amniotic extracallular matrix, ②hat were related to amniotic membrane and amniotic extracellular matrix as the scaffold materials in tissue engineering, and ③that were related to the amniotic cells as the seed cells in tissue engineering were selected. The irrelevant and repeated articles were excluded.
DATA EXTRACTION: Totally 90 articles related to the application of amniotic membrane and amniotic extracellular matrix in tissue engineering were collected and 30 met the inclusive criteria.
DATA SYNTHESIS: ① Normal human amniotic membrane consists of epithelial cell layer, basement membrane, compact layer, fibrocyte layer and spongy layer. The epithelial call layer contains amniotic epithelial cells and various growth factors secreted by them. Fresh amniotic extracellular matrix has the similar compact layer to the extracellular matrix preserved for middle and long terms. ②Amniotic membrane and amniotic extracellular matrix could be used as the repair materials in peripheral nerve tissue engineering, skin tissue engineering, corneal tissue engineering, tendon tissue engineering, and bone tissue engineering. ③Amniotic calls are composed of amniotic epithelial cells, flbroblasts, and undifferentiated mesenchymal calls. ④The related questions and future direction of the application of amniotic membrane and amniotic extracallular matrix in tissue engineering are also analyzed.
CONCLUSION: Human amniotic membrane and amniotic extracallular matrix as a kind of biomaterial should be sampled, preserved and used strictly according to the medical standard, but it has not been unified so far. With the development of biotechnique and other cross-correlated technique, the application of amniotic membrane and amniotic extracallular matrix in tissue engineering would have a prosperous future.
