摘要
目的探讨多球壳菌素(ISP-Ⅰ)对体外激活的大鼠肾小球系膜细胞(GMC)的调控作用。方法分别采用MTT法及流式细胞术检测IL-1、IL-6对GMC的激活作用及ISP-Ⅰ对激活后GMC增生的抑制作用。结果MTT法结果显示10U/mlIL-1及1000U/mlIL-6刺激GMC24、48h后,反映细胞增生程度的吸光度(A)值较对照组增高(P<0.05);激活的GMC在25~500μg/mlISP-Ⅰ作用24、48h后的A值较对照组低(P<0.05)。流式细胞术检测的结果与MTT法一致,即GMC被激活12、24、36h时的细胞增生率高于对照组(P<0.05);激活的GMC在100μg/mlISP-Ⅰ作用下,细胞增生率低于对照组(P<0.05)。结论ISP-Ⅰ对激活后的GMC有明显抑制作用。
Objective To study the effects of myriocin (ISP-Ⅰ) on activated glomerular mesangial cells (GMCs) in rats. Methods MTT method and FCM were used to determine proliferation rate of GMCs stimulated by IL-1, IL-6 and the anti-pro- liferation rate of ISP-Ⅰ on activated GMCs. Results The absorbance (A) value, which represented the cell proliferation rate, of GMCs stimulated by IL-1 and IL-6 was higher than in control group. The A value of activated GMCs inhibited by ISP-Ⅰ was lower than in GMCs activated by IL-1, IL-6; FCM revealed the percentages of proliferative GMCs stimulated by IL-1, IL-6 were higher than those in the controls. FCM also indicated the percentages of proliferative cells in ISP-Ⅰ treated group were lower than in those stimulated by IL-1, IL-6. Conclusion ISP-Ⅰ can significantly inhibit the activated GMCs.
