摘要
目的:探讨低浓度α-生育酚对神经元细胞膜的保护作用.方法:培养SD胎鼠皮层神经元细胞作为实验对象.将神经元细胞进行分组:正常对照组、氧自由基损伤组和不同浓度α-生育酚处理组(10,20,40和80mg/L).用Fenton反应对神经元细胞膜造成损伤.在损伤后1及2h分别对各组神经元进行PI染色,利用激光共聚焦显微镜观察神经元细胞膜的损伤情况,并用TBA法检测神经元细胞外液中丙二醛(MDA)生成量.结果:80mg/L浓度α-生育酚可减少氧自由基对神经元细胞膜的损伤,且可减少MDA生成量.结论:80mg/L浓度α-生育酚可以有效对抗氧自由基对神经元细胞膜的损伤.
AIM: To evaluate the protective effect of α- tocopherol at a low concentration on neuronal membrane. METHODS: The neurons from the cortex of the SD embryonic mice were divided into several groups: normal control group, oxidative damage group, and α-tocopherol treatment groups (10, 20, 40 and 80 mg/L). The oxidative damage was induced in oxidative damage group and α-tocopherol treatment groups by Fenton reaction. The neurons were stained with PI 1 and 2 h after the oxidative damage for observing the condition of the cell membrane of the neurons with the confocal laser scanning microscope. The malondialdehyde (MDA) levels were detected in the extra cellular solution of different groups by TBA method. RESULTS : 80 mg/ L α-tocopherol reduced the damage of the oxidative free radicals to the cell membrane of neurons and the production of MDA. CONCLUSION: 80 mg/L α-toeopherol can protect the cell membrane of neurons from the damage of oxidative free radicals effectively.
