姜黄素对大鼠环孢素慢性肾毒性防治作用研究 被引量:5

Prevention of Cyclosporine A(CSA)-induced Chronic Nephrotoxicity with Curcumin in Rats

摘要 目的观察姜黄素对环孢素(CSA)肾毒性的防治作用。方法将30只大鼠随机分为4组,即正常组6只,模型组、治疗组和预防组各8只。正常组给予橄榄油30 mg.kg-1.d-1灌胃4周;模型组以CSA30 mg.kg-1.d-1灌胃4周;治疗组以CSA30 mg.kg-1.d-1灌胃2周后,再给予姜黄素200 mg.kg-1.d-1灌胃2周;预防组给予CSA30mg.kg-1.d-1与姜黄素200 mg.kg-1.d-1联合灌胃4周。观察给药前后各组大鼠的体重变化,以及给药后24 h尿量、血肌酐、尿素氮和肾脏病理变化。用免疫组织化学和RT-PCR法检测各组大鼠肾组织HO-1和bFGF表达情况。结果与正常组相比,模型组大鼠给予CSA后体重下降(P<0.05),尿量增多(P<0.05),尿素氮、肌酐上升(均P<0.05),肾脏组织出现较典型的病理变化,HO-1表达显著下降(P<0.01),bFGF表达增加(P<0.01)。与模型组相比,治疗组和预防组HO-1表达增加(均P<0.01),bFGF表达下降(均P<0.01)。结论姜黄素可改善CSA所导致的肾毒性,可能是通过增加HO-1、降低bFGF的表达发挥抗CSA肾毒性的作用。 Objective To evaluate the effect of curcumin on cyclosporine A (CSA)-induced nephrotoxicity. Methods 30 rats were randomized into g groups, which included model group, curcumin - treated group and curcumin - prevented group with 8 rats in each, as well as control group with 6 rats. The rat stomach was perfused by olive oil 30 mg·kg^-1·d^-1 for g weeks in control group, CSA 30 mg ~ kg - t . d - l for g weeks in model group, CSA 30 mg·kg^-1·d^-1 for 2 weeks followed by curcumin 200mg·kg^-1·d^-1for another 2 weeks in curcumin - treated group, and CSA 30 mg·kg^-1·d^-1 combined with curcumin 200 mg·kg^-1·d^-1 for g weeks in curcumin - prevented group. The weight before and after CSA medication, the urinary volume[ mL· (24 h)^-1), serum cretinine, BUN and renal pathological changes after CSA application were recorded. The expression of HO - land bFGF in rat kidney was examined by immunohisochemistry and RT - PCR. Results In comparison with control group, the results in rat model were as follows : the weight was decreased ( P 〈 0. 05 ), urinary volume was increased( P 〈 0. 05 ), BUN and CR were elevated( P 〈 0. 05 ). Renal pathological study displayed the typical changes of CSA nephrotoxicity. The expression of HO-1 were decreased obviously ( P 〈0. 01 ) and the expression of bFGF was increased( P 〈0. 01 ). In comparison with model group, HO-1 expression was increased ( P 〈 0. 01 ) but bFGF expression was decreased (P 〈0. 01 ) in curcumin-treated and-prevented groups. Conclusion Curcumin could counteract CSA-induced nephrotoxicity, probably through up-regulation of HO-1 expression and down - regulation of bFGF expression in the kidney.
