摘要
目的研究APP17肽对血管性痴呆大鼠行为学及额叶Bcl-2,Bax蛋白表达的影响。方法采用双侧颈总动脉结扎方法制备前脑缺血致血管性痴呆大鼠模型,并观察APP17肽的保护作用,采用Morris水迷宫检测行为学,免疫组织化学检测额叶Bcl-2,Bax蛋白表达。结果手术组与对照组相比认知能力明显下降(P<0.01),Bcl-2、Bax蛋白表达增加(P<0.01),治疗组与手术组相比认知能力改善(P<0.01),Bcl-2蛋白表达明显增加(P<0.01),Bax蛋白表达明显减少(P<0.01)。结论APP17肽可以通过调节Bcl-2、Bax蛋白表达来抑制细胞凋亡,改善血管性痴呆大鼠的认知能力。
Objectlve To study the Effects of APP17-mer pepfide against the expression of Bcl-2 and Bax protein in frontal lobe of Vascular Dementia Rats. Methods VD model was formed by adopting the permanent occlusion of bilateral common carotid arteries in Wistar rats to produce the forebrain ischemia. And the protective effect of APP17-mer peptide was observed.cognitive ability of rats were studied with the Morris water maze . The expression of Bcl-2 and Bax protein in frontal lobe were studied with immunohistochemieal staining. Results VD medal tnay decrease cognitive ability( P 〈 0.01), increase the expression of Bcl-2 and Bax protein in frontal lobe( P 〈 0.01), APP17 -mer peptide significant alleviated cognitive ability( P 〈 0.01 ) , markedly increase the expression Bcl-2 protein( P 〈 0.01), and reduce expression of Bax protein( P 〈 0.01). Conclusion APP17 -mer pepfide can inhibit apoptosis through regulating the expression of Bcl-2 and Bax protein in frontal lobe. APP17 -mer pepfide may improve cognitive ability in Vascular Dementia rats.
