摘要
目的:研究番石榴叶提取物对大鼠结肠炎组织损伤的保护作用及其机制.方法:用健康Wistar大鼠建立大鼠结肠炎模型,灌胃用药2wk后,评价大鼠结肠黏膜损伤指数(CMDI),检测结肠组织髓过氧化物酶(MPO)及超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量和细胞肿瘤因子(TNF)、白细胞介素-8(IL-8)、白细胞介素-10(IL-10)水平.结果:不同剂量番石榴叶提取物(200,500,800mg/kg)灌胃均能不同程度降低模型大鼠CMDI(2.62±0.47,2.06±0.54,1.83±0.43vs3.15±0.39,P<0.05,P<0.01,P<0.01),MPO活性(2240.78±479.26,2078.42±377.91,1748.85±236.71nkat/gvs2695.37±624.79nkat/gP<0.05,P<0.01,P<0.01),减少MDA含量(2.16±0.41,1.97±0.35,1.96±0.30,P<0.05,P<0.01,P<0.01),提高SOD活性(3618.06±538.44,3756.92±577.45,4197.51±375.74nkat/gvs2663.37±603.79,P<0.05,P<0.01,P<0.01),降低TNF(2.65±0.40,2.51±0.47,2.21±0.41μg/Lvs3.30±0.63μg/L,P<0.05,P<0.01,P<0.01)、IL-8(1.20±0.33,1.05±0.24,0.92±0.13μg/Lvs1.53±0.38μg/L,P<0.05,P<0.01,P<0.01)、IL-10(39.36±9.65,50.26±11.32,59.68±13.65ng/Lvs30.74±12.91ng/L,P<0.05,P<0.01,P<0.01)水平,且与用药剂量呈一定量效关系.结论:番石榴叶提取物通过拮抗氧化、免疫调节、损伤修复作用缓解结肠炎大鼠炎症反应,减轻结肠损伤.
AIM: To explore the protective effect of guava leaf water extract on the colonic tissues with ulcerative colonitis induced by trinitrobenzene sulfonic acid (TNBS) as well as its mechanism in rats.
METHODS: Rat model of ulcerative colonitis was established by clyster of TNBS. After treatment with guava leaf extract for 2 weeks, the colon mucosal damage index (CMDI) was evaluated. The activity of myeloperoxidase (MPO), superoxide dismutase (SOD), the content of malondialdehyde (MDA) in colon tissues, and the levels of tumor necrosis factor (TNF), interleukin-8 (IL-8) and interleukin-10 (IL-10) in serum were examined.
RESULTS: Guava leaf extract (200, 500, 800 mg/kg) decreased the values of CMDI (2.62± 0.47, 2.06 ± 0.54, 1.83 ± 0.43 vs 3.15 ± 0.39; P 〈 0.05, P 〈 0.01, P 〈 0.01), the activity of MPO (2240.78 ± 479.26, 2078.42 ± 377.91, 1748.85 ± 236.71 nkat/g vs 2695.37 ±624.79 nkat/g; P 〈 0.05, P 〈 0.01, P 〈 0.01), the contents of MDA (2.16 ± 0.41, 1.97 ± 0.35, 1.96 ± 0.30 μmol/g vs 2.66 ± 0.65 μmol/g; P 〈 0.05, P 〈 0.01, P 〈 0.01), and the levels of TNF (2.65 ± 0.40, 2.51 ± 0.47, 2.21 ± 0.41 μg/L vs 3.30 ± 0.63 μg/L; P 〈 0.05, P 〈 0.01, P 〈 0.01), IL-8 (1.20 ± 0.33, 1.05 ± 0.24, 0.92 ± 0.13 μg/L vs 1.53 ± 0.38 μg/L, P 〈 0.05, P 〈 0.01, P 〈 0.01) and IL-10 (39.36 ± 9.65, 50.26 ± 11.32, 59.68 ± 13.65 ng/L vs 30.74 ± 12.91 ng/L, P 〈 0.05, P 〈 0.01, P 〈 0.01), but increased the activity of SOD (3618.06 ± 538.44, 3756.92 ± 577.45, 4197.51 ± 375.74 nkat/g vs 2663.37 ± 603.79 nkat/g, P 〈 0.05, P 〈 0.01, P 〈 0.01) in a dose-depended manner.
CONCLUSION: Guava leaf extract can relieve the inflammatory reaction and colonic injury in colonitis rats by immunoregulation, anti-oxidation and wound repair.
