摘要
研究了特非那丁对映体在含有疏水性L-酒石酸酯的1,2二氯乙烷溶液及甲醇水溶液两相中的苇取分配行为,考察了不同烷基链长的L-酒石酸酯,L-酒石酸酯的浓度,有机溶剂的种类,及其溶解特非那丁的甲醇水溶液浓度对分配系数K和分离因子α的影响。实验表明:L-酒石酸酯与特非那丁Ⅱ对映体形成的复合物稳定性比与特非那丁Ⅰ对映体形成的复合物稳定性要大,三类有机溶剂的萃取性能为醇〉1,2二氯乙烷〉烷烃:甲醇的浓度在50%~90%时,随着溶解特非那丁的甲醇水溶液中的甲醇浓度的增加,分配系数和分离网子均降低:当甲醇的浓度为50%时,可以得到最佳的K和α;随着L-酒石酸酯的浓度的提高,分配系数K和d分离因子先增加然后减小;当L-酒石酸酯的浓度约为0.25mol/L时,K和α达到最大值;L-酒石酸酯的碳链长度对分配系数K和分离因子α也有很大的影响。
The distribution of terfenadine enantiomers was examined in the methanol aqueous and organic solvent of two phase system containing a chiral selector L-tartric esters . And the influence of the length of alkyl chain of L-tartric esters, the concentration of L-tartric esters, organic solvents and the concentration of metha- nol aqueous on the partition coefficients (K) and separation factors (α) of terfenadine, was invesgated. The L-tartric esters formed more stable complexes with Ⅱ-terfenadine than that of Ⅰ-terfenadine. The extraction performance of the three kinds of organic solvents is different, e. i. , alcohols 〉 1,2-dichloroethane 〉 alkyl compounds. When the concentration of the methanol in the methanol aqueous is 50% - 90%, with increasing the concentration of the methanol, the partition coefficients and separation factors decrease, K and a are at the best when the concentration of methanol is 50%. With the increase of concentration of L-tartfic esters, K and ct increase firstly, then decrease. And K and a are at the best when the concentration of L-tartfic esters is 0. 225mol/L. The length of alkyl chain of L-tartic esters also has great influence on K and α.
