摘要
目的:探讨VEGFC、VEGFD在乳腺癌组织中的表达及其作用。方法:采用免疫组织化学方法对112例乳腺癌组织切片染色,观察乳腺癌组织中VEGFC,VEGFD以及LYVE1的表达情况。结果:112例乳腺癌组织中,VEGFC阳性率为83.0%,VEGFD阳性率为75.9%。二者表达呈正相关(r=0.767,P<0.001)。VEGFC阳性指数在淋巴结转移组(60.54±16.73)明显高于未转移组(43.56±15.64)(P<0.05)。VEGFD阳性指数在淋巴结转移组(60.42±16.26)明显高于未转移组(41.82±15.93)(P<0.05)。随着癌细胞VEGFC和VEGFD表达强度增强,LYVE1阳性淋巴管数也随之增加,各组间均有显著性差异(P<0.001)。VEGFC和VEGFD的表达与LYVE1阳性淋巴管均呈一定相关性,(前者r=0.864,P<0.001;后者r=0.870,P<0.001)。乳腺癌中LYVE1阳性淋巴管数在淋巴结转移组(13.72±7.74)明显高于未转移组(7.69±5.85)。结论:VEGFC及VEGFD在人乳腺癌组织呈高水平表达,表达强度与肿瘤间质淋巴管密度及肿瘤淋巴结转移有关;肿瘤间质淋巴管密度与肿瘤淋巴结转移密切相关。
Objective:To identify the expressions of vascular endothelial growth factor (VEGF)-C and VEGF-D in primary breast cancer and their significance. Methods:A series of 112 primary breast cancer specimens were detected for the expressions of VEGF-C,VEGF-D as well as lymphatic vessel endothelial hyaluronan receptor (LYVE)-1 by immunohistochemical method. Results:Among 112 cases of breast cancer, the positive rates of VEGF-C and VEGF-D were 83.0% (93/112) and 75.9% (85/ 112), respectively. The expression of VEGF-C and VEGF-D had positive correlation (r=0. 767 ,P〈0. 001). The positive index of VEGF-C and VEGF-D in breast cancer with lymph node metastasis was significantly higher than that with no lymph node metastasis[(60.54±16.73) vs (43.56±15.64),P〈0.05; (60.42±16.26) vs (41.82±15.93),P〈0.05]. With the increase in the expression of VEGF-C or VEGF-D, the number of LYVE-1 positive vessels also increased. There was significant difference between each group(P〈0. 001). Expression of VEGF-C or VEGF-D was correlated with the number of LYVE-1 positive vessels,respectively (r=0. 864,P〈0. 001;r=0. 870, P〈0. 001). The number of LYVE-1 positive vessels in breast cancer with lymph node metastasis was significantly higher than that without lymph node metastasis [ ( 13. 72 ± 7. 74) vs ( 7. 69 ± 5. 85 )]. Conclusion:VEGF-C and VEGF-D are over expressed in primary breast cancer. Their expressions are related to the number of LYVE-1 positive vessels and lymph node metastasis. The number of LYVE-1 positive vessels is closely related to lymph node metastasis.
