Effects of hyperbaric oxygen on the cell cycle of colorectal carcinoma LoVo cells

摘要 目的探讨高压氧(HBO)对结肠癌LoVo细胞周期的影响。方法应用流式细胞仪检测不同压力及时间高压氧暴露后LoVo细胞的细胞周期改变。结果①0.2 MPa HBO可使LoVo细胞在细胞周期S期积聚,与0.15、0.25.MPa HBO暴露组比较,差异具有极显著性(P<0.01);②0.2 MPa HBO暴露后24 h组S期细胞积聚明显高于HBO暴露后12 h和48 h组(P<O.01);③0.2MPa HBO暴露后24 h组的S期细胞积聚高于其他组别(P<0.01)。结论高压氧可诱导肿瘤细胞进入细胞周期并在S期细胞积聚,细胞周期同步化提示高压氧可作为大肠肿瘤放疗和化疗的辅助治疗。 Objective To investigate the effects of hyperbaric oxygen on the cell cycle of colorectal carcinoma LoVo cells. Methods Cell cycle change of LoVo cells exposed to HBO at different pressures and for different durations was examined with flow cytometry. Results 0. 2 MPa HBO exerted the highest cell accumulating effect, compared with other pressures (0. 15 MPa,0.25 MPa, P 〈 0. 01 ), and the effect was the strongest at 24 h duration ( P 〈 0.01 ). The best effect was exerted by HBO at 0.2 MPa for 24 h. Conclusions HBO could induce tumor ceils to enter the cell cycle and accumulate at S stage,which suggested that HBO was an useful adjuvant therapy for tumor cell chemotherapy or radiation in the treatment of large bowel neoplasm.
