摘要
采用紫外光谱、红外光谱、核磁共振、原子力显微镜观察等现代分析手段以及特异性β-葡聚糖酶水解,研究了燕麦β-葡聚糖提取物组分POG-1A的结构特性。结果表明,POG-1A是由D-吡喃葡萄糖残基通过β-(1→3)和β-(1→4)糖苷键连接成的线性均一多糖,其中β-(1→3)和β-(1→4)键的比例为1∶2.4;特异性β-葡聚糖酶水解后主要的酶解产物为纤维三糖和纤维四糖,它们占90.91%;原子力显微镜显示POG-1A的高级结构为复杂的网络状结构,酶解以后的产物为链长不等的聚集物。
A series of modern analysis methods, such as UV, IR, NMR and AFM (atomic force microscope) as well as specific β-glucanase hydrolysis were used to analyze the structure features of an oat β--glucan component, POG - 1 A. The result shows that POG - 1 A is a linear, non - branch polysaccharide containing a single type ofβ- - D - glucose. D - glicopyranose linked with β- - (1→β-) and β-- (1→4) glucosidic bonds. The ratio of β- - ( 1→3 ) linkage and β- ( 1→4 ) linkage is 1 : 2.4. The oligosaccharides produced byβ- - glucanohydrolase mainly are cellotriose and cellotetrose. The total amount of tri - and tetra- saccharide is 90.91%. We observed the association of POG - 1 A directly using AFM. As showing in the result, POG - 1A is composed of cross - linked network struc- ture and the structure afterβ- - D - glucan - 4 - glucanohydrolase hydrolysis is polymerized by irregular fragments.
