摘要
目的利用复发性口疮(recurrent oral ulcertion,ROU)动物模型,探讨T淋巴细胞与ROU发病机制的关系及观察银杏叶提取物(Extract Ginkgo biloba,EGb)对ROU外周血T淋巴细胞的影响及治疗作用。方法用大鼠口腔粘膜匀浆组织加弗氏免疫佐剂注射方法建立大鼠ROU模型,将模型动物分为阳性对照组、阴性对照组、EGb糊剂组、EGb片剂组、EGb糊剂+片剂组。观察用药前后溃疡数目、直径、持续时间、间歇期的变化;应用流式细胞仪检测用药前后CD4+细胞、CD8+细胞的变化。结果①用药后EGb糊剂组、EGb片剂组、EGb糊剂+片剂组、阳性对照组的ROU溃疡数目减少、直径缩小、持续时间缩短、间歇期明显延长(P<0.01),EGb糊剂组与阳性对照组之间没有差异,其中EGb片剂组的溃疡数目、直径、间歇期值与阳性对照组比较有显著差异(P<0.01),EGb糊剂+片剂组的溃疡持续时间、间歇期值与阳性对照组相比有显著差异(P<0.01),EGb片剂组与EGb糊剂+片剂组相比对溃疡的数目减少、直径缩小、间歇期延长作用没有差异(P>0.05);②用药后EGb糊剂组、EGb片剂组、EGb糊剂+片剂组、阳性对照组的CD4+细胞升高(P<0.05),及CD4+/CD8+比值升高(P<0.05),其中EGb片剂组与EGb糊剂+片剂组之间没有差异(P>0.05),与阳性组之间也没有差异(P>0.05)。结论ROU的发生与T淋巴细胞失调有关,EGb能够调节大鼠的免疫功能,对ROU有较好的治疗作用,尤以EGb片剂、EGb糊剂+片剂效果最好。
Objective Using the rat's recurrent oral ulcertion (ROU) models and to study directly the pathogenesis and the effects of the Extract Ginkgo biloba(EGb) on the recurrent phthous ROU and to explore effect on peripheraic blood lymphocyte the effective plant amedica drug for cure ROU. Methods Establishing ROU models by injecting the mixture of homogenate tissue of rat's oral mucosal and immunologic adjuvant into 50 SD rats. The rats were randomly divided into 5 groups, including Positive group, Negative group, EGb cataplasm + troche group , EGb troche group, and EGb cataplasm group. The changes of CD4^ + , CDs^+ in blood were observed using flow - cytometre before and after the therapy. Results In the cataplasm + troche group, the troche group, the cataplasm group, and the positive group, the CD4^+ and the ratio of CD4^+ / CDs^ + were obviously increased after the medication, There was no difference among the troche group, the cataplasm + troche group and the positive group. Conclusion EGb could increase CD4^ + and CD4^+ / CDs^ + ,decrease the virulence of cell, and adjust immunity of rats, especially the cataplasm + troche and the troche were the most effective.
