基本培养基和6-BA对花生去子叶胚离体培养形态发生的影响 被引量:4

Effects of basic media and 6-BA on morphogenesis from decotyledon peanut (Arachis hypogaea L.) embryo during in vitro culture

摘要 将已去子叶的粤优1号花生(Arachis hypogaeaL.)的胚、胚芽、上胚轴、下胚轴、胚轴作外植体在MS、1/2 MS和B5培养基上进行离体培养,结果表明,B5对愈伤组织的诱导和萌发成苗的效果最好,外植体中以胚轴诱导愈伤组织最好,成苗诱导以胚最好.在B5+0.20 mg/L NAA上,6-BA含量为0.20 mg/L时胚萌发的幼苗生长最好,根、茎的POD活性及木质素含量也较高,含量太低或偏高均不利于幼苗的形态发生. The explants such as embryos, embryonic axes, embryonic buds, epicotyls, hypocotyls from decotyledons 'Yue you No.1' peanut (Arachis hypogaea L. ) were in vitro cultured. Among MS, 1/2 MS and B5 media, the 135 medium was the best for callus inducing and seedling growing in all conditions. Among other explants, embryo was the best for callus inducing. Embryo was more effective than embryonic bud for seedling growing. The higher peroxidase (POD) activity, and higher hgnin content of buds, the best effect for seedling growing from embryo, were all obtained by the using of B5 medium with the addition of 0.20 mg/L NAA and 0.20 mg/L 6-BA.
