四逆汤类方提取物对离体小鼠腹腔巨噬细胞免疫功能的影响 被引量:18

Effect of the Extracts from Sini Tang Serial Decoctions on the Immune Function of Mouse Peritoneal Macrophages in vitro

摘要 目的:探讨四逆汤类方提取物对小鼠腹腔巨噬细胞免疫功能的影响。方法:小鼠腹腔巨噬细胞经不同浓度的四逆汤类方提取物作用后,中性红法检测吞噬能力。以巨噬细胞培养液上清对L929细胞和胸腺淋巴细胞增殖的影响为指标,MTT比色法检测肿瘤坏死因子(TNF)和白细胞介素-1(IL-1)的生成量。结果:附子、干姜配葱白提取物(EAGO),附子、干姜配甘草提取物(EAGL),附子、干姜、葱白配胆汁提取物(EAGB),附子、干姜、甘草配人参提取物(EAGG)可以提高小鼠腹腔巨噬细胞的吞噬功能,但与附子配干姜提取物(EAG)组比较无显著差异。与模型对照组比较,EAGO、EAGL显著减少TNF生成量,EAGL和EAGG可以显著降低IL-1生成量。结论:四逆汤类方均可以提高腹腔巨噬细胞吞噬功能,EAGL和EAGO可以降低TNF生成量,EAGL和EAGG可以降低IL-1的生成量。对LPS诱导炎症反应的实验模型,EAGL的药理活性较全面。类方之间对腹腔巨噬细胞免疫功能的影响不尽相同。 Objective:To explore effect of the extracts from Sini Tang Serial Decoctions(ESTSD)on the immune function of mouse peritoneal macrophages. Methods: After the cultured mouse peritoneal macrophages treated with different concentrations(25,5, lgg/mL)of ESTSD,the phagocytotic activity was determined by neutral red method, the productions of interleukin-1 (IL-1)and tumor necrosis factor (TNF) from the supernatant were determined by MTT colorimetry based on the proliferation of L929 and thymic lymphocyte respectively. Results: Compared with the model control group, our results indicated that ESTSD including extracts of Aconite root, Dried ginger and Fistular onion stalk(EAGO)and extracts of Aconite root, Dried ginger and Licorice root (EAGL), extracts of Aconite root, Dried ginger, Fistular onion stalk and pig' s bile ( EAGB ), extracts of Aconite root, Dried ginger, Licorice root and ginseng (EAGG) increased significantly the phagocytotic activity. The supernatant of peritoneal macrophages treated with EAGL and EAGO decreased significantly the production of TNF compared with model group. The supernatant of peritoneal macrophages treated with EAGL and EAGG decreased significantly the production of IL-1 compared with model group. Conclusion:The results suggested that ESTSD stimulated the phagocytotic activity and EAGL, EAGG decreased the production of IL-1 as well as EAGL, EAGO decreased the production of TNF. EAGL had all pharmacologic effect on mouse peritoneal macrophages in this model. Obviously, the effect of ESTSD on these parameters of mouse peritoneal macrophages were different in character.
