摘要
采用一步法和两步法对黄牛皮进行预处理以提取保留了天然结构的胶原,在比较了它们对黄牛皮的脱毛效果、皮纤维的分散情况以及天然胶原提取率的基础上,选用两步处理法对黄牛皮进行预处理。考察两步法中过氧化氢浓度、作用时间、pH值对皮胶原纤维分散的影响,所处理的皮采用酶法提取天然胶原时提取率较高。采用电泳及DSC分析了所得天然胶原的分子质量和变性温度,结果表明:所得天然胶原相对分子质量集中,其变性温度为38.5℃,为保持了三股螺旋结构的天然胶原。
To extract collagen with the natural structure from bovine hide, pretreatments with one-step and two step methods were studied. Based on the unhairing effects, collagen fiber dispersion and collagen extraction percentage, the two- step pretreatment method was chosen. The effects of hydrogen peroxide concentration, time and pH on collagen fiber dispersion were analyzed. Natural collagen could be extracted at high percentage from bovine hides pretreated with two step method. SDSPAGE electrophoresis and DSC were used to analyze collagen molecular weight distribution and denaturation temperature resixectively. Results indicated that the molecular weight was concentrative and the denaturation temperature was 38.5°C, showing triple helix structure of natural collagen.
