摘要
目的:制备一种普通光学显微镜下可见的细胞标记用大粒径彩色高分子微球。方法:以苯乙烯及丙烯酰胺为原料,在体系中加入电解质,有机溶剂及油溶性染料,采用无乳化剂乳液聚合法制备彩色高分子微球,并通过酰肼化和羧化两步反应在微球表面衍生出羧基。检测了微球直径和微球表面的羧基含量,讨论了各种聚合条件对微球直径的影响。结果:微球直径为3士0.05m,色泽鲜艳,表面羧基含量为190.5mol/g(干固体).
Objective: To prepare a kind of large colored microspheres which can be observed under optical microscope and used for cell labelling. Methods: In the presence of organic solvent, electrolyte and dye, microspheres [styrene/acrylamide/dye (St AAm Dye) copolymer] were synthesized from styrene and acrylamide by means of emulsifier free emulsion polymerization. The carboxyl groups on the surface were derived from acrylamide by hydrazinolysis and carboxylate reaction. The diameters of microspheres and the content of carboxyl groups on the surface have been measured. The effect of the polymerization condition on microsphere size was discussed. Results: The diameter of the colored microsphere was 3±0.05 μm; the content of carboxyl groups on the surface was 190.5 μmol/g (dry solids). Conclusion: The results indicate that monodisperse and colored large polymer microspheres can be obtained by these reported methods.
