摘要
目的:从苦瓜中分离水溶性苦瓜多糖,并研究其组成性质。方法:采用DEAE纤维素柱层析和凝胶柱层析分离纯化得到苦瓜多糖MCP2和MCP4,SepharoseCL-6B凝胶柱层析和醋酸纤维膜电泳检测证明为均一组分。气相色谱法和薄层层析法测定其多糖组成,凝胶过滤法测定其相对分子质量,红外光谱测定其糖苷键类型。结果:苦瓜多糖MCP2为半乳聚糖,MCP4含有鼠李糖、木糖、半乳糖和阿拉伯糖,它们的摩尔比为1:0.75:1.83:0.85。平均相对分子质量分别为5.37×104,9.65×104。它们的红外光谱中有典型的多糖吸收峰,紫外扫描无核酸和蛋白特征吸收峰。结论:MCP2,MCP4首次从苦瓜中提取获得,为均一多糖组分。
Objective: To study the isolation, purification and composition of Momordica charantia L. polysaccharide(MC). Methods MC was purified by DEAE ion-exchange cellulose (DEAE-52) and Sephadex G-100.The components of the saccharide were determined by GC and TLC. Its MW was measured by gel filtration. The form of glucosidic bond was determined by IR. Results: Rhamnose, xylose galactose, and arabinose were found in MCP4 with the molar ratio of 1:0.75:1.83:0.85 while galactose was found in MCP2, Their average MWs were estimated respectively to be 5.37 × 10^4 and 9.65 ± 10^4. Typical absorption of polysaccharide was shown in their IR spectrum. They had no typical absorption of nucleic acid or protein by UV scanning. Conclusion: MCP2 and MCP4 are first isolated from Momordica charantia L.
