摘要
根据GenBank数据库的单核细胞增生李斯特菌iap基因设计2对引物,建立了套式PCR快速检测单增李斯特菌的方法。两对引物分别扩增出约1 500 bp和500 bp片段,与预期大小一致。套式PCR方法灵敏性实验检测极限为10 cfu/mL;人工污染猪肉检测极限为103cfu/mL;整个检测过程可在7 h内完成。采用该法对南海口岸进口的冻肉进行检测,检测出9份阳性,与传统的细菌分离方法完全一致,说明套式PCR方法具有很好的特异性。
According to the sequence data published by Genbank, two pairs of primers were designed to amplify the iap gene of Listeria monocytogens and a Nested polymerase chain reaction(Nested-PCR) method for the detection of Listeria monocytogens has been developed. Two pairs of primers amplified 1 500 bp and 500 bp fragment respectively as expected. The detection limit is 101 cfu/mL, while the detection limit is 10^3 cfu/mL when detecting the artificially contaminated swine meat. The whole detection can be finished in 7 hours. 9 listeria monocytogens were detected from the import frozen meat, which is accorded with that of the ISO11290-1 method.
