藏药金腰草中黄酮诱导K562细胞凋亡及其分子机制研究 被引量:7

K562 Apoptosis Induced by Flavone from Tibetan Medicine Chrysosplenium Nudicaule Bunge and its Molecular Mechanism

摘要 目的研究藏药金腰草中6,7,3′-三甲氧基-3,5,4′-三羟基黄酮对人红白血病细胞株K562的凋亡诱导效应及其可能的作用机制。方法采用四氮唑蓝(MTT)比色法、透射电镜、细胞周期分析法和DNA琼脂糖凝胶电泳检测K562细胞凋亡;采用流式细胞术(FCM)测定细胞Bcl-2、Fas蛋白表达水平。结果6,7,3′-三甲氧基-3,5,4′-三羟基黄酮可以抑制K562细胞增殖,并呈现典型的凋亡形态改变;细胞周期分析显示S期细胞数明显增多,出现S期阻滞;DNA琼脂糖凝胶电泳可见明显的梯状条带;6,7,3′-三甲氧基-3,5,4′-三羟基黄酮作用72 h后,Bcl-2蛋白阳性表达率由36.10%下降到27.78%,Fas蛋白阳性表达率由11.33%上升到32.76%。结论6,7,3′-三甲氧基-3,5,4′-三羟基黄酮对K562细胞有抑制增殖和诱导凋亡作用。其诱导凋亡可能与下调Bcl-2和上调Fas有关。 Objective To study the apoptosis of K562 cells induced by 6,7,3'-trimethoxy-3,5,4'-trioxy-flavone from Tibetan Medicine Chrysosplenium nudicaule Bunge and the possible mechanism. Methods MTT colorimetric assay,electronmicroscope, cell cycle analysis, and DNA agarose gel electrophoresis were used to examine apoptosis in K562 cells. Expression levels of Bcl-2 and Fas protein were measured with flow cytometry. Results 6,7,3'-trimethoxy-3,5,4'-trioxy-flavone inhibited proliferation of K562 cells. Typical morphologic features of apoptosis were observed. Cell cycle analysis indicated increased S phase proportion,as well as apparent blockage of S phase. Agarose gel electrophoresis showed evident DNA fragmentation. After treated by 6,7,3'-trimethoxy-3,5,4'- trioxy-flavone for 72 h,the Bcl-2 positive cells decreased from 36.10% to 27.78% and the Fas positive cells increased from 11.33% to 32.76 %. Conclusion 6,7,3'-trimethoxy-3,5,4'-trioxy-flavone can inhibit proliferation and induce apoptosis of K562 cells, whose mechanisms may be carried out via down-regulatiop the expression of Bcl-2 and up-regulation the expression of Fas.
