摘要
目的:探讨黄芪在急性肺损伤(AL I)时对肝细胞生长因子(HGF)表达的影响,评价黄芪在AL I中的治疗机制。方法:制备内毒素(LPS)性AL I模型。设正常对照组(生理盐水1 m l/kg)、模型组(LPS 5 m g/kg)、黄芪治疗组(黄芪8 m g/kg)。用逆转录聚合酶链反应(RT PCR)检测各组大鼠肺组织匀浆中HGF mRNA表达的变化;用免疫组化法检测各组大鼠肺组织中HGF阳性细胞数的变化。结果:RT PCR检测结果显示,AL I时HGF mRNA表达明显增加(P<0.05),黄芪能促使HGF mRNA表达进一步升高(P<0.05)。免疫组化显示,AL I时HGF阳性细胞表达产物增加(P<0.01),黄芪能促进HGF的表达(P<0.05)。结论:黄芪通过促进HGF的表达来促进AL I的修复。
Objective: To investigate the effect of astragalus on expression of hepatocyte growth factor (HGF) in the acute lung injury (ALI) and the mechanism of astragulus in the treatment of ALI. Methods: Twenty-four rats were randomly divided into three groups: normal control group (1 ml/kg normal saline), ALI group (5 mg/kg lipopolysaccharide) and astragulus treatment group (8 mg/kg astragulus). HGF mRNA in lung homogenate was detected by reverse transcription- polymerase chain reaction (RT- PCR). Expression of HGF protein was observed by immunohistochemistry. Results: The level of HGF mRNA and expression of HGF protein in the lung tissue of ALI increased, compared with that of the normal lung tissue(P〈0.05). After treatment of astragalus, the expression of HGF mRNA and HGF protein increased further (P〈0.05). Conclusion: Astragulus can repair the injured lung tissue by inducing the expression of HGF.
