Direct Assay of Cephalexin in Human Plasma by High Performance Capillary Zone Electrophoresis

摘要 报道了采用高效毛细管区带电泳技术,直接将人血浆注入毛细管中进行人血浆中头孢氨苄含量测定的新方法。试验表明,以磷酸盐(pH9.67)作缓冲溶液,对人血浆中头孢氨苄进行直接分析具有较高的灵敏度和较好的重复性。 A direct assay of cephalexin in human plasma by high performance capillary zone electrophoresis (HPCZE) is described. The sample, untreated human plasma, was directly introduced into the capillary and a phosphate buffer solution(pH 9. 67) was employed. The capillary used was 47cm×75μm i. d. and detection was carried out with a UV monitor at 254 nm. The calibration curve showed a good linearity(correlation coefficient =0. 9988)in the concentration range of 5-80μg/ml. for cephalexin. The total time for separation and determination was within 10 min. This method is characterized by direct assay of sample in plasma without any pretreatment. The results showed that HPCZE is a simple, rapid, sensitive and reliable assay method.
