摘要
目的探讨牛磺酸(Tau)对阿霉素(Adr)心脏毒性与心肌一氧化氮合酶(NOS)表达的影响。方法将26只新西兰纯种兔随机分成3组(对照组、阿霉素组和牛磺酸治疗组),用药后测定3组动物的心输出量(CO)、左心室收缩压(LVSP)和左心室舒张末压(LVEDP),并取心肌组织作HE染色及组化染色进行图像定量分析。结果与对照组比较,阿霉素组CO、LVSP显著降低,LVEDP显著增高(均P<0.01);经牛磺酸治疗后,治疗组CO和LVEDP显著回升或下降(均P<0.01);阿霉素组和牛磺酸治疗组的心肌NOS光密度(OD)值均较对照组显著升高(P<0.01),而治疗组又较阿霉素组显著升高(P<0.01)。结论阿霉素能使心肌组织NOS表达增强及释放NO的量增加,参与心肌损伤过程;牛磺酸对阿霉素导致的心肌损伤有保护作用。
Objective To investigate the effect of Taurine on heart toxicity induced by Adriamycin and the expression of Nitric oxide synthase. Methods The CO,LVSP.LVEDP of the control,Adr and Adr+Tau group were measured among three groups of 26 New Zealand rabbits. HE staining and histochemical method were used in the myocardium and the image analysis was also performed. Results In the Adr group, the CO, LVSP were significantly decreased and the LVEDP were significantly increased (P〈0.01) compared with the control group. In the Adr+Tau group,the CO,LVEDP were significantly improved (P〈0.01)compared with the Adr group. The image analysis showed which the optical density(OD) values of NOS in the myocardium there were significantly improved (P〈0.01).Conclusion Tau could protect Adr-induced damage of the myocardium. Adr increased the expression of NOS in the myocardium and increased the production of NO and participate in the progress of damage of the myocardium.
