甲状腺肿瘤组织中p14^(ARF)基因纯合性缺失及其产物表达状况的研究 被引量:4

摘要 目的:检测甲状腺肿瘤组织中p14ARF基因变异及其产物的表达,探讨此基因与甲状腺肿瘤发生发展的关系。方法:应用双重PCR和免疫组化SP法分别检测20例甲状腺腺瘤和28例甲状腺癌组织中p14ARF基因纯合性缺失及其蛋白表达状况。28例甲状腺癌包括11例甲状腺滤泡癌,12例乳头状癌,4例髓样癌以及1例未分化癌。结果:1)p14ARF基因在甲状腺腺瘤及癌中的纯合性缺失率分别为5.0%(1/20)和42.9%(12/28),两者差异有统计学意义,P=0.0036。2)p14ARF蛋白在甲状腺腺瘤及甲状腺癌中的阳性率分别为90.0%(18/20)和35.7%(10/28),两者差异有统计学意义,P=0.0002。结论:甲状腺肿瘤中p14ARF基因的纯合性缺失是其蛋白表达失活的重要机制之一,与甲状腺肿瘤的发生发展密切相关。p14ARF可能是甲状腺肿瘤的分子标志物之一,可作为鉴别甲状腺腺瘤与滤泡癌的辅助指标。 OBJECTIVE: To study the relationship between gene p14^ARF and thyroid tumors by measuring alteration of gene p14^ARF and expression of its product METHODS: Twenty thyroid adenomas and 28 thyroid carcinomas (11 folicular carcinomas. 12 papillary carcinomas, 4 medullary carcinomas and 1 undifferentiated carcinoma) were assayed for alteration of p14^ARF and expression of its product, including homozygous deletion, by differential PCR, and protein expression by immunohistochemistry. RESULTS: 1) Homozygous deletion rates of p14^ARF in thyroid adenomas and carcinomas were 5. 0% (1/20), 42.9% (12 28) respectively,and there was a significant difference between adenomas and carcinomas. P=0.003 6. 2)Positive rates of protein p14^ARF in thyroid adenomas and carcinomas were 90.0%, 35.7% respectively, and there also was a significant difference between adenomas and carcinomas, P=0. 000 2. CONCLUSIONS: In thyroid tumors, homozygous deletion of p14^ARF is one of important mechanisms of loss of protein and is related to the happening and development of thyroid tumors, p14^ARF may be one of molecular markers of thyroid tumors and can be used as an assistant index to distinguish thyroid folicular carcinoma from adenoma.
