抗人心肌肌球蛋自重链单克隆抗体的筛选与鉴定 被引量:1

Production and Characterization of Monoclonal Antibody Directed against Human Cardiac Myosin Heavy Chain

摘要 用自制人心肌肌球蛋白作为哺抗原,按杂交瘤常规技术获得一株稳定的G4杂交瘤细胞株。单抗亚型为IgG1型;用免疫印迹法对成人、胎儿、哺乳类动物及鲫鱼的心肌肌球蛋白提取液在200kD处有一着色带;免疫组化ABC法表明单抗与成人、胎儿、猪、兔、大白鼠的心肌细胞胞浆均有阳性反应,与血管平滑肌均呈阴性反应。该单克隆抗体可用于与心肌坏死病理过程有关的疾病的诊断。 Balb/c mice were immunized by injection of human cardiac myosin extracted by ourselves. By the routine hybridoma techniques, a stable G4 specific hybridoma cell line was obtained. This monoclonal antibody was detected to be IgG1. Western blot analysis for antigen detection using specific anti-myosin heavy chain McAb showed a band at 200kD position which included human, fetus and other animal cardiac myosins. Immunohistochemistry studies with ABC method indicated that left ventricular specimens from above cardiac tissues showed cardiac cytoplasms in which myosin existed were positively stained. In contrast, arterial blood vessels appeared to be negatively stained. The results suggested that this monoclonal antibody may be used for th diagnosis of those diseases related to the pathologic courses of myocardial necrosis.
