摘要
目的:观察不同时限内神经肽Y(NPY)对心肌细胞胞浆及核内游离钙浓度的作用。方法:用Fluo3AM荧光标记细胞内游离钙离子,用NPY(100nmol/L)刺激WISTAR乳鼠心肌细胞,检测0min、5min、10min及24h内心肌细胞胞浆及核内钙离子浓度变化。结果:加入100nmol/LNPY即刻至10min内,心肌细胞胞浆及核内钙含量差异均无显著性。经100nmol/LNPY刺激24h后,心肌细胞胞浆钙含量(16.81±2.1)明显高于对照组(7.45±1.96)(P<0.01),核内钙含量(30.65±4.06)也显著高于对照组(15.64±2.16)(P<0.01)。结论:较长时间的NPY刺激可导致心肌细胞胞浆及核内钙浓度增加。
Objective:To investigate the effect of neuropeptide Y(NPY) on [Ca^2+] in plasma and nuclear of cardiomyocytes during various period. Method: Cardiomyocytes of neonatal Wistar rats were cultured with NPY (100 nmol/L) for 0 min, 5 min, 10 min and 24 h. Fluorescent assay with Fluo3-AM was used to detect [Ca^2+] in plasma and nuclear of cardiomyocytes. Result: Within 10 min,there were no considerable changes of the [Ca^2+] in plasma and nuclear stimulated by NPY(100 nmol/L). 24 hours after incubation, the [Ca^2+] in plasma (16.81±2.1)was significantly elevated compared with control group(7.45±1.96)(P<0.01), and the [Ca^2+] in nuclear(30.65±4.06)was also distinctly increased compared with control group(15.64±2.16)(P<0.01). Conclusion: NPY caused the enhancement of [Ca^2+] in plasma and nuclear of cardiomyocytes in rats.
