摘要
目的研究小剂量甲氨蝶呤(MTX)治疗异位妊娠的药动学。方法12例异位妊娠患者肌注MTX20mg。于给药前和给药后0.17,0.5,1,2,3,4,6,8,12和24h抽取静脉血,采用荧光偏振免疫分析法(FPIA)测定MTX血药浓度。用3P97软件计算其药动学参数。结果小剂量肌注MTX符合开放二室动力学模型。结论此研究结果有助于甲氨蝶呤合理应用,为临床用药提供理论依据。
OBJECTIVE: To investigate the pharmacokinetic parameters of low-dose methotrexate in the treatment of ectopic pregnancy. METHODS: After a single intramuscular admintration of methotrexate 20 mg was given to 12 patients with ectopic pregnancy, the serum samples were collected from 0 to 24 h. MTX concentration in serum was measured by fluorescence polarization immunoassay(FPIA). The pharmacokinetic parameter were calculated with 3P97 program software. RESULTS: The pharmacokinetic profile of low-dose methotrexate in patients with ectopic pregnancy was described by a two-compartment open model. CONCLUSION: These results may be helpful to a rational individualized methodtrexate dosage regimen.
