体外冲击波碎石术致肾细胞内钙水平与氧自由基的变化 被引量:1

The varieties of intracellular calcium ion level and oxygen free radicals in the kidney of rabbit following extracorporeal shock wave lithotripsy

摘要 目的:观察体外冲击波碎石术(E SW L)对肾细胞内游离钙(犤C a2犦i)水平和肾组织氧自由基的影响及+钙通道阻滞剂的干预作用,探讨肾损伤机制。方法:30只家兔制成单肾模型,随机等分为对照组、E SW L组和E SW L+维拉帕米预处理组。E SW L后24h获取肾脏,采用Fura-2/A M荧光指示剂测定肾细胞内犤C a2+犦i浓度变化,同时测定肾组织超氧化物歧化酶(SO D)活性和脂质过氧化终产物丙二醛M D A。结果:ESW L后肾细胞内犤C a2+犦i增高,肾组()织SO D活性降低、M D A生成增加(P<0.01);维拉帕米预处理能抑制细胞内犤C a2犦i和M D A增加(P<0.01,P<+0.05),提高SO D活性P<0.01)。结论:E SW L所致肾细胞内C a2+犦i超载和肾组织氧自由基侵袭,可能在E SW L肾(犤损害病理过程中起重要作用。 Objective To observe the varieties of intracellular calcium ion (i) and oxygen free radicals in the kidney of rabbit following extracorporeal shock wave lithotripsy (ESWL) and effects of pretreatment with calcium channel blocker on these varieties in order to study the mechanism of renal injuries induced by ESWL. Methods Thirty healthy male domestic rabbits were surgically managed to the mono nephron models and randomized into 3 groups consisting of ten each randomly: control group, ESWL group and ESWL plus verapamil pretreatment group. The rabbits were sacrificed 24 hours after ESWL. Renal intracellular i level was detected by fura 2/AM fluorescence assay, superoxide dismutase (SOD) and malonic aldehyde (MDA) in the renal tissue were measured at the same time. Results Compared with control group, intracellulari level and MDA content increased significantly (P< 0.01), SOD activity reduced prominently (P< 0.01) in ESWL group. Intracellular i level and MDA content obviously decreased(P< 0.01, P< 0.05 ), SOD activity increased strikingly(P< 0.01) in ESWL plus verapamil pretreatment group compared with those in ESWL group. Conclusions Renal intracellulari overloading and oxygen free radicals invasiveness might have an important role in pathologic process of renal injuries induced by ESWL.
