益气散结泄水复方中配伍温热药对癌性胸水小鼠生存期及胸水中相关免疫细胞因子的影响 被引量:2

Effects of yiqi sanjie xieshui complex prescription combined with hot and warm herb on the survival time of the malignant pleural effusion mice model and the immunity-related cytokine in pleural effusion

摘要 目的:探讨在益气散结泄水复方中配伍温热药对癌性胸水模型小鼠生存期的影响及对胸水中白细胞介素2(IL-2)、γ干扰素含量的影响。方法:实验于2003-09/12在湖南中医学院完成。在用艾氏腹水癌细胞瘤株对昆明种小白鼠进行胸腔注射的方法复制癌性胸水模型72只,随机分为:益气散结泄水复方组(简称复方组),复方+低温热组,复方+高温热组,温热药组,环磷酰胺对照组(简称化疗组),荷瘤对照组(简称模型组),给药9d后计算各组平均生存天数及生命延长率。相同方法造模、分组、给药,9d后断头处死小鼠,ELISA法测胸水中IL-2、γ干扰素含量。结果:复方+高温热组、复方+低温热组、复方组、化疗组小鼠生存期分别为(19.3±1.3),(19.0±2.0),(16.9±2.2),(14.2±2.9)d,模型组为(11.8±1.9)d,差异有显著性意义(P<0.01);复方+高温热组、复方+低温热组小鼠生存期较复方组显著延长,差异有显著性意义(P<0.05);单用温热组小鼠生存期为(11.2±1.9)d,未见延长。复方+高温热组、复方+低温热组胸水中γ干扰素含量为(16.23±2.81),(16.67±2.55)ng/L,较复方组犤(14.60±2.89)ng/L犦显著增加,差异有显著性意义(P<0.01);复方加高温热组胸水中IL-2含量为(129.50±21.13)ng/L,较复方组犤(113.41±15.82)ng/L犦增加,差异有显著性意义(P<0.05); AIM:To investigate the effect of yiqi sanji xieshui(complex prescription) combined with hot and warm herb on the survival time of malignant pleural effusion mice model and the levels of interleukin 2(IL 2),interferon gama(IFN γ ) in pleural effusion. METHODS:The experiment was conducted in the Hunan College of Traditional Chinese Medicine from September to December 2003.EAC cell suspension was intrapleurally infused into Kunming mice to duplicate 72 malignant pleural effusion mice models.The models were randomized into yiqi sanjie xieshui complex prescription group(complex prescription group),complex prescription combined with low dose warm and hot herb group,complex prescription combined with high dose warm and hot herb group,hot and warm herb group,cyclophosphamide control group(chemotherapy group) and tumor bearing control group(model group).The mean survival time and life prolonged rate of all the models were calculated 9 days after drug administration.The methods of modeling,grouping and administration were used in the same way.All mice were sacrificed 9 days after drug administration.The levels of IL 2 and IFN γ in pleural effusion were measured by ELISA. RESULTS:The survival time was significantly longer in complex prescription combined with high or low dose hot and warm herb group,complex prescription group and chemotherapy group[(19.3± 1.3),(19.0± 2.0),(16.9± 2.2) and (14.2± 2.9) days respectively] than in model group[(11.8± 1.9) days](P< 0.01),and also significantly longer in complex prescription combined with high or low dose warm and hot herb group than in complex prescription group(P< 0.05),but it was not prolonged in hot and warm herb group[(11.2± 1.9) days].The level of IFN γ in pleural effusion of complex prescription combined with high or low dose warm and hot herb group[(16.23± 2.81) and(16.67± 2.55) ng/L] were significantly higher than that in complex prescription group[(14.60± 2.89) ng/L](P< 0.01).The level of IL 2 was significantly higher that in complex prescription combined with high dose warm and hot herb group[(129.50± 21.13) ng/L] than that in complex prescription group[(113.41± 15.82) ng/L](P< 0.05).The levels of IFN γ and IL 2 in warm and hot herb group were higher than those in model group,but there were no significant difference between them. CONCLUSION:Combined with warm and hot herb,yiqi sanjie xieshui complex prescription can prolong the survival time and increase the levels of IFN γ and IL 2 in pleural effusion of malignant pleural effusion mice model,but the complex prescription used only can not.
