摘要
以大花君子兰(Clivia miniata)的幼嫩子房组织为外植体,在附加2mg/LKT和lmg/L2,4-D的MS基本培养基上培养,诱导愈伤组织的频率可达90%以上,诱导出的愈伤组织经同一诱导培养基长期继代培养后可以“一次成苗”。胚性愈伤组织在2~6年的继代培养过程中,其增殖生长与苗的分化同时进行,分化频率随着继代培养的延长而上升,经长期继代培养后,已建立了具有持久高分化潜力的胚性愈伤组织无性系,由无性系不断分化产生的大量再生植株,移栽到盆钵中能够成活,并且生长良好。
The calli are induced from unpollinated ovaries of Clivia miniata cultured on the MS medium containing 2mg/L 6-BA and 1mg/L 2, 4-D. a great deal of regenerative plants are obtained through subculturing for long time in same mediam. The differentiation frequency of calli has been increased following to 2~3 years. It is found that the callus possesses high ability of differentiation.
