摘要 目的:研究甘草总黄酮对超氧阴离子和羟自由基的清除和抑制作用。方法:用化学反应法检测不同剂量的甘草总黄酮对黄嘌呤一黄嘌呤氧化酶系统产生的超氧阴离子和Fenton反应产生的羟自由基的清除作用,并与维生素E和甘露醇的抗氧化作用进行比较。结果:甘草总黄酮具有明显清除氧自由基的作用,其半数清除量(IC50)为226mg/L明显低于维生素E的IC50770mg/L,清除氧自由基的作用明显优于维生素E(P<0.05);甘草总黄酮对Fenton反应生成的羟自由基具有较强的直接清除作用,该作用明显优于甘露醇,其清除羟自由基的IC50是甘露醇的1/255,其抑制羟自由基生成的IC50是甘露醇的1/139。结论:甘草总黄酮对氧自由基的羟自由基有清除和抑制作用,且作用强于维生素E和甘露醇。 AIM: To investigate the effects of total flavonoids from radix glycyrrhizae in scavenging and inhabiting superoxide anion and hydroxyl free radical. <METHODS: The chemical reaction method was used to examine the scavenging effect of total flavonoids from radix glycyrrhizae on superoxide anion, produced by xanthine/xanthine oxidase system, and hydroxyl radical, produced by Fenton reaction, and the results were compared with the anti oxidation of vitamin E and manical.<RESULTS: Total flavonoids from radix glycyrrhizae had an obvious effect in scavenging superoxide anion, its inhibitory concentration of 50% (IC50) was 226 mg/L, obviously lower than that of vitamin E(770 mg/L). Its effect in eliminating oxygen derived free radicals was obviously superior to that of vitamin E(P< 0.05). Total flavonoids from radix glycyrrhizae had stronger effect in eliminating the hydroxyl radical produced by Fenton reaction directly, which was significantly superior to that of manicol, and its IC50 to hydroxyl radical was 1/255 of manicol,and its inhibition on hydroxyl radical from generating IC50 was 1/139 of manicol.<CONCLUSION: Total flavonoids from radix glycyrrhizae has the effect of eliminating and inhibiting oxygen derived free radicals and hydroxyl radical, which are superior to those of vitamin E and manicol.
