摘要
目的 :研究烟碱依赖对大鼠脑皮层c -fos表达的影响。方法 :采用异硫氰酸胍法提取大鼠脑皮层区总mRNA ,经Dotblot,Northernblot杂交技术检测c-fos基因在大鼠脑皮层的表达。结果 :烟碱 (Nic)急性给药组c -fosmRNA的表达水平与NS组比较差异有显著性 (P <0 0 1) ;烟碱依赖组c -fosmRNA表达水平明显高于Nic急性给药组 (P <0 0 1)。结论 :烟碱依赖大鼠脑皮层c-fos的表达增强。
Objective: To study the effect of nicotine dependence on c-fos gene expression of cortex in rats. Methods: Single-step method for RNA isolation by acid guanidinium-isothiocyanate-phenol -chloroform extraction and Dot blot, Northern blot techniques were used to detect the expression of c-fos gene in cortex in rats. Results: Acute nicotine treatment group caused a rapid and marked induction of c-fos mRNA level in cortex(P<0.01), and the c-fos mRNA level of nicotine dependence group was higher than that of acute nicotine treatment group(P<0.01). Conclusion: Nicotine dependence can increase the c-fos gene expression of cortex in rats.
