摘要
以具有不同O和H抗原的沙门氏菌制备免疫原,用常规方法产生杂交瘤细胞,经过对不同O和H抗原沙门氏菌全或无反应的筛选,得到4株反应谱广的沙门氏菌单抗,分别命名为CB_8、de_7,、cc_6和DD_4。单抗CB_8和de_7在对沙门氏菌属的反应性上具有互补的特点,二者可和受试的A-O_(67)群沙门氏菌及亚利桑那沙门氏菌83个血清型共96株中的95株发生反应,覆盖率达99%,其中包括了在我国引起人类疾病和食物中毒的绝大多数菌型,但在属外不与受试的21株其他肠杆菌反应,从而为沙门氏菌检验提供了优良试剂。同时,这组单抗在沙门氏菌共同抗原和致病机理的研究方面亦具有重要意义。
Four hybridoma cell lines secreting monoclonal antibodies (MAbs) with specificity to common antigenic epitopes shared by different serotypes of Salmonella spp., designated as CB_8, de_7, ce_6 and DD_4 respectively, were derived from separate fusions with SP2/0 myeloma cells and spleen cells of Salmonella immunized BABL/c mice. Two of these MAhs, CB_8 and de_7, complemented each other in terms of binding reactivities with different serotypcs of the genus Salmonella. CB_8 and de_7 strongly re acted with 95 out of 96 known Salmonella strains belonging to 83 serotypes, a total of 99% coverage as shown in ELISA, in contrast, definite negative reactions were found with other 21 bacterial strains in the family of Enterobacteriaceae including Escherichia, Shigella, Citrobacter, Enterobacter, Klebsiella, Proteus, Yersinia, and Serratia. The only one Salmonella strain which did not react with MAbs CB_8 and de_7 was found to be reactive with MAbs Vi-6-6 developed in our laboratory earlier. The other two MAbs, cc_6 and DD_4, reacted with 92 and 78 Sallmonella strains respectively while they also reacted with a few of other Enterobacterial strains in the same assay. All the results suggested that the MAbs would be very useful in the analysis and localization of common antigenic epitopes shared by different serotypes of Salmonella and in the development of diagnostic kit for rapid detection of Salmonella sop. in food, feed and clinical specimens.
