老年慢性阻塞性肺疾病气道黏蛋白修饰性基因表达的改变 被引量:1

摘要 目的 了解老年慢性阻塞性肺疾病 (COPD)气道黏液特性改变的分子基础。方法 采用原位杂交技术检测老年正常肺组织和老年COPD患者肺组织中对黏蛋白分别起糖基化和硫酸化修饰作用的β 1 ,4半乳糖苷转移酶和N 乙酰氨基葡萄糖 6 0 硫转移酶的转录水平。结果 老年COPD患者肺组织中上述糖转移酶和硫转移酶转录水平较正常肺组织明显升高 ,在缓解期已分别升高了约 4倍和 3 5倍 ,而在急性发作期则约升高 6倍和 5倍。 Objective To investigate the molecular basis of the changes in the properties of airway mucin in elder patients with chronic obstructive pulmonary disease (COPD).Methods The transcriptional levels of β-1,4-Galactosyl transferase (β-1,4-GT) and N-Acetylglucomaine-6-O-Sulfotransferase (GLcNAc-6-O-ST) by which the airway mucin was glycosylated and sulfated respectively were detected with in situ hybridization technique in the lung tissue of normal elders and clinic remission stage. Results β-1,4-GT and GLcNAc-6-O-ST mRNA expression of lung tissue in elder patients with COPD were both significantly higher than that of normal lung tissue. The levels of the expression were enhanced about 4 times and 3.5 times of the normal levels respectively in remission stage of COPD and were increased about 6 times and 5 times of the normal in the acute attack stage. Conclusions The high transcriptional levels of β-1,4-GT and GLcNAc-6-O-ST in the lung tissue of elder patients COPD could be important molecular basis of intensification of the glycosylated and sulfated degree of the airway hypersecretory mucin.
