摘要
目的研究pp’-DDE对离体培养大鼠睾丸支持细胞转铁蛋白表达的影响。方法对大鼠睾丸支持细胞离体原代培养,运用一步法RT-PCR检测给与pp’-DDE后24h支持细胞转铁蛋白mRNA水平。结果睾丸支持细胞转铁蛋白mRNA水平随pp’-DDE所给剂量的增高而降低,且与对照组差异存在显著性(P<0.05),呈剂量依赖关系。结论pp’-DDE可抑制睾丸支持细胞转铁蛋白的基因转录,从而抑制其合成,导致精子发生障碍和生殖功能紊乱。
Objective To explore the effect of p,p'-DDE on transferrin expression in Sertoli cells of rat testis in vitro. Methods The effects of p,p'-DDE on expression of mRNA of transferrin gene in primary cultured Sertoli cells exposed to p,p'-DDE for 24 h was examined by one-step RT-PCR methods. Results The transferrin mRNA levels were decreased with the increases of p,p'-DDE concentration in Sertoli cell cultures,and there was a significant dose-effect relationship(P<0.05). Conclusion p,p'-DDE could inhibit the translation of transferrin in rat Sertoli cells cultured in vitro.
