摘要
分别采用包埋玻璃化法、玻璃化法、简化玻璃化法、一般低温保存法对枣疯病愈伤组织、枣疯病组培苗茎尖及带腋芽茎段、健康婆枣组培苗茎尖及带腋芽茎段、长春花组培苗茎尖及茎段、长春花栽培苗茎尖及茎段进行了低温(-20、-76、-196℃)保存研究。结果表明,长春花只有组培苗茎尖在液氮(-196℃)中保存48 h后恢复培养成功,而其他温度(-20、-76℃)、其他材料均不能成功恢复培养;所有枣树材料低温保存后进行恢复培养时均死亡。
By using encapsulation-vitrfication,vitrification,simplified vitrification and common preservation,the preservation methods of the stem apex and stem with axillary buds of tissue culture seedling with symptom of jujube witche’s broom,healthy jujube,Catharanthus roseus and callus of jujube witche’s broom seedlings at-20 ℃,-76 ℃,-196 ℃.The result showed that the stem apex of tissue culture seedling of C.roseus was successfully recovered in liquid nitrogen(-196 ℃) for 48 h;the other preservation methods of C.roseus at-20 ℃ and-76 ℃ and cryopreservation of jujube materials did not succeed.
