雏鸵鸟脾脏的显微与超微结构及脾内5-HT的分布定位 被引量:4

摘要 【目的】观察雏鸵鸟脾脏的解剖学、显微和超微结构特点,探明5-HT免疫反应阳性细胞在脾内的分布规律。【方法】采用光镜和电镜制片及染色技术,并结合免疫组化SABC法对5-HT在脾脏内的表达进行定位。【结果】雏鸵鸟脾脏被膜较薄,小梁不发达;白髓内动脉周围淋巴鞘薄,偶见脾小结,但无生发中心;淋巴细胞内含较多的线粒体、游离核糖体及少量RER,核膜双层,有明显的核膜孔;椭球体积小,其周围淋巴鞘较厚,且数量多,椭球与周围淋巴组织之间有强嗜酸性均质物形成的明显界限,鞘毛细血管外细胞胞质内富含RER、线粒体和游离核糖体。5-HT免疫反应阳性细胞呈圆形或卵圆形,胞体较大,胞质较多,核多位于细胞一侧,阳性产物位于胞质内;红髓内阳性细胞数量最多,阳性产物表达最强;动脉周围淋巴鞘内阳性细胞数量较少,但阳性产物表达较强;而椭球内有弱阳性产物表达。【结论】雏鸵鸟椭球周围淋巴鞘数量远比动脉周围淋巴鞘的多;椭球与周围淋巴组织之间界限明显,其间的强嗜酸性物质主要由纤维构成;5-HT免疫阳性细胞数量较多,主要位于红髓、动脉周围淋巴鞘和椭球,阳性产物表达强;脾脏是合成外周5-HT的重要场所,可能主要由浆细胞和原浆细胞合成分泌;外周5-HT可能参与机体的体液免疫应答反应。 【Objective】The experiment was to study the anatomy traits, microstructure and ultra-structure of spleen in ostrich chicks. The distribution of 5-HT immunoreactivity (5-HTir) positive cells was also investigated.【Method】Light and transmission electron microscopical methods were used to observe the morphological structure of ostrich spleen. The location of 5-HT in the spleen was studied with immunohistochemical method, SABC.【Result】The spleen capsule in ostrich chicks was thin, and peritrabecular was underdeveloped. The periarterial lymphatic sheath in white pulp was thin. There were splenic nodules occasionally, no germinal center was observed. The lymphocyte with rich mitochondria, free ribosomes and RER in the cytoplast had double membrane and obvious nuclear pores. The bulk of ellipsoid was small, but the lymphatic sheath around it was thick and the number of it was plenty. There was obvious demarcation line which was composed of acidophilic matter between the ellipsoids and the peripheral lymphatic tissue. The cells around sheathed capillary were rich in RER, mitochondria and ribosome in the cytoplasm. 5-HTir positive cells were round or oval,whose bulk was large, cytoplast was rich and nucleus migrated. The positive products located in the cytoplast. The number of positive cells in the red pulp was the most, and the expression was the strongest. The number of positive cells in the periaterial lymphatic sheath was less, but the expression was stronger. The expression in the ellipsoids was weak.【Conclusion】The number of periellipsoidal lymphatic sheath is far more than that of the periaterial lymphatic sheath in the ostrich chicks. The demarcation line between ellipsoids and peripheral lymphatic tissue is obvious, and the acidophilic matter is mainly made up of fiber. The number of 5-HTir positive cells is abundant, which mainly locate in the red pulp, periaterial lymphaticsheath and ellipsoids. The expression of positive products is stronger. The spleen is the important location which produces the peripheral 5-HT, and the peripheral 5-HT may be biosynthesized by plasma cells and the original plasma cells. The peripheral 5-HT may be related to the anamnestic response of humoral immunity in the ostrich chicks.
