摘要
目的:研究对慢性应激抑郁模型大鼠海马BDNF表达与认知功能损伤之间的关系。方法采用21d慢性不可预见的温和应激处理结合孤养制备抑郁大鼠模型。运用被动躲避实验评定大鼠的学习记忆能力,采用荧光定量PCR技术和免疫组织化学技术检测BDNF基因和蛋白的表达变化。结果抑郁组大鼠与正常对照组比较,体重、糖水消耗量、旷场试验运动均减少,被动规避缺陷,以上行为学改变差异均有统计学意义(P<0.05)。免疫组织化学和实时荧光定量PCR技术检测均显示:抑郁组大鼠海马各区BDNF蛋白和mRNA相对表达量均显著低于正常对照组,差异有统计学意义(P<0.01)。结论慢性应激抑郁大鼠海马区BDNF表达下降可使认知障碍加重。
Objectives To make clear of the relationship between brain-derived neurotrophic factor(BDNF)expression and cognitive impairment in depression rats.Methods Isolated-living condition and chronic unpredictable mild stress are applied successively to set up depression rat model.Learning and memory function is evaluated by passive avoidance test.Real Time-PCR and immunohistochemistry method are applied to detect the BDNF mRNA and BDNF protein.Results Compared with the normal control group,the depression group have significant changes(P<0.05):weight loss,the decrease of sucrose solution consumption,the drop of open-field behavior,the deficiency of passive avoid test.Compared with the normal control group,BDNF expression is significantly decreased in the hippocampus of the depression group(P<0.01).Conclusions The decreased expression of BDNF in the hippocampus of depression rats may make the cognitive impairment increase.
