瑶山甜茶通过调节线粒体自噬改善糖尿病大鼠并发症的实验研究 被引量:5

The Study of Rubus suavissimus S. Lee (RS) Improving the Diabetic Complications of the Streptozotocin (STZ) Rats by Promoting Mitophagy

摘要 目的: 研究瑶山甜茶 (RS) 对链脲佐菌素 (STZ) 致糖尿病大鼠并发症改善的作用机制.方法: 用STZ腹腔注射建立糖尿病模型, 并随机分为模型组、甜茶高剂量组、甜茶低剂量组、阳性药组, 测定各组大鼠心、肝、肾的脏器指数, 并取心肌、肾脏和肝脏组织行透射电镜检测.结果: 甜茶高、低剂量组的心、肝、肾的脏器指数较模型组显著降低 (P<0. 01或P<0. 05), 各脏器超微结构趋向修复, 凋亡减少, 并且出现线粒体自噬现象.结论: 瑶山甜茶可以显著降低糖尿病大鼠的血糖, 保护各脏器功能, 其机制可能为其有效活性成分可诱导线粒体自噬, 抑制凋亡, 从而改善各脏器功能, 线粒体自噬有可能为瑶山甜茶改善糖尿病并发症的新靶点. objective To study the mechanism of Rubus suavissimus S.Lee(RS)which could alleviat diabetic complications in the Streptozotocin(STZ)rats.Method Rats had been induced by 55 mg/kg STZ.gastric administration and became diabetic model rats.They were randomly divided into model group,high dose group,low dose group and positive group.The ultrastructure and the organ index of the hearts,livers and kidneys of the rats were detected.Results Compared with the model group,the organ index of the livers,hearts,kidneys was decreased significantly in the high and low does groups(P<0.01 or P<0.05),and the ultrastructures of each organ were restored,besides,there has been less apoptosis and more mitophagy.Conclusion RS can significantly reduce blood glucose in diabetic rats and help to maintain the organ function.The mechanism is likely that the effective constituent of RS could promote mitophagy while suppress apoptosis in order to alleviate diabetic complications.Mitophagy is possible to be a new treatment target for RS to alleviate diabetic complications.
