摘要
目的:分析原发性肝癌患者的血清肿瘤标志物与其肿瘤组织当中凋亡调节基因的表达。方法:选取本院2017-08~2018-08收治的50例原发性肝癌患者以及同期接受健康体检的50例健康者作为研究对象,分别建立研究组与对照组,检测2组研究对象的血清肿瘤标志物GP73、TK1以及DKK1表达水平,分析肿瘤组织当中的凋亡调节基因表达情况。结果:研究组患者的血清中GP73、TK1以及DKK1表达水平相比于对照组均明显更高,对比差异存在统计意义(P<0.05);肝癌组织Livin、Plk1与Xiap等抑制凋亡基因的表达水平与癌旁正常组织相比均明显更高(P<0.05),而肝癌组织Caspase-3、Caspase-8与MTS1等促凋亡基因的表达水平与癌旁正常组织相比均明显更低(P<0.05);血清GP-73、TK1以及DKK1表达水平均与肝脏组织的抑制凋亡基因表达水平存在正相关关系,与促凋亡基因的表达存在负相关关系。结论:对于原发性肝癌患者血清的肿瘤标志物TK1、GP-73以及DKK1表达水平会出现异常升高,血清TK1、GP73、DKK1表达水平均与肝脏组织当中凋亡抑制基因表达呈正相关,且与促凋亡基因表达呈负相关,检测肿瘤标志物是有效判断原发性肝癌病情情况的指标。
Objective:To analyze the relationship between serum tumor markers in patients with primary liver cancer and the expression of apoptosis-regulating genes in tumor tissues.Methods:From August 2017 to August 2018 were 50 cases of patients with primary liver cancer (research group) and have a medical examination during the same period of 50 cases of healthy volunteers (control group) as the research object, the detection of two groups of serum tumor markers GP73, TK1 and DKK1 expression level, analysis of apoptosis regulating gene expression in tumor tissues.Results:The serum levels of GP73, TK1 and DKK1 were higher in the study group, and the difference between the two groups was statistically significant (P<0.05). The expression levels of apoptosis inhibiting genes such as Livin, Plk1 and Xiap in liver cancer tissues were significantly higher than those in normal adjacent tissues (P<0.05), while the expression levels of pro-apoptotic genes such as caspase-3, caspase-8 and MTS1 in liver cancer tissues were significantly lower than those in normal adjacent tissues (P<0.05). The expression levels of serum gp-73, TK1 and DKK1 were positively correlated with the expression levels of apoptotic inhibiting genes in liver tissues, and negatively correlated with the expression levels of pro-apoptotic genes.Conclusion:The expression levels of tumor markers TK1, gp-73 and DKK1 in serum of patients with primary liver cancer are abnormally increased, and positively correlated with the expression of apoptotic inhibiting genes in liver tissues, and negatively correlated with the expression of pro-apoptotic genes.
