摘要
目的 探讨miRNA在肝癌中的表达差异,建立其与肝癌预后的关系.方法 提取TCGA数据库中的肝癌样本的miRNA表达量及肝癌临床数据,根据正常组织与肿瘤组织分为两组,通过edgeR算法对比两组数据的miRNA表达量.联合差异miRNA与临床生存时间作生存分析,寻找对肝癌预后产生影响的miRNA.结果 肝癌组织对比正常组织共发现247 个差异表达miRNA (P<0. 01),通过生存分析发现17个差异miRNA与预后相关(P<0. 05),Cox多因素分析结果提示miRNA-30d、miRNA-939、miRNA-188、miRNA-5003与肝癌患者生存时间相关(P<0. 05),差异均有统计学意义.结论 miRNA-30d、miRNA-939、miRNA-188、miRNA-5003在肝肿瘤组织对比正常组织表达差异且肝癌预后相关,对miRNA在肝癌领域的研究具有借鉴价值.
To investigate the differential expression of miRNAs in hepatocellular car-cinoma and to establish a correlation between differential expression of miRNA and prognosis of liver canc-er.Methods The miRNA expression of liver cancer samples and clinical data of liver cancer were extrac-ted from TCGA database.According to the normal tissue and tumor tissue,the miRNA expression levels of the two groups were compared by the edgeR algorithm.Survival analysis was performed by combining differ-ential miRNAs with clinical survival time to find miRNAs that have an impact on the prognosis of liver cancer.Results A total of 247 differentially expressed miRNAs were found in HCC tissues compared with normal tissues(P<0.01).Survival analysis revealed that 17 differential miRNAs were associated with prog-nosis(P<0.05).Cox multivariate analysis revealed miRNA-30d and miRNA-939.miRNA-188 and miR-NA-5003 were associated with survival time of patients with liver cancer(P<0.05),and the difference was statistically significant.Conclusion miRNA-30d,miRNA-939,miRNA-188 and miRNA-5003 are differen-tially expressed in liver tumor tissues compared with normal tissues and prognosis of liver cancer.It is useful for the study of miRNA in the field of liver cancer.
