响应面法优化西藏猫乳总黄酮提取工艺及其抗氧化活性研究 被引量:7

Study on optimization of extraction technology and antioxidant activity of the total flavonoids from Rhamnella gilgitica Mansf. et Melch by response surface method

摘要 目的:采用响应面法优化西藏猫乳总黄酮提取工艺,并对其抗氧化能力进行考察。方法:利用超声法提取西藏猫乳总黄酮;选取提取温度、提取时间、乙醇浓度、料液比进行单因素实验,在单因素实验基础上,运用Box-Behnken试验设计,以总黄酮提取率为响应值,优化提取工艺;应用DPPH、ABTS及超氧自由基清除等试验,对超声法和回流法所得产物抗氧化能力进行比较。结果:西藏猫乳总黄酮的最优提取工艺参数:料液比为1∶48.42mL/g,提取时间50.00min,乙醇浓度53.33%,温度54.66℃,总黄酮提取率为8.16%。西藏猫乳总黄酮具有良好的抗氧化作用,且超声提取法抗氧化能力优于回流提取法。结论:超声提取法可充分提取西藏猫乳总黄酮,且提取产物具有良好的抗氧化作用。 Objective:To optimize the extraction process of total flavonoids from Rhamnella gilgitica Mansf.et Melch by the response surface method,and to investigate its antioxidant capacity.Methods:Total flavonoids from Rhamnella gilgitica Mansf.et Melch were extracted by ultrasonic method.The extraction temperature,extraction time,ethanol concentration and solid-liquid ratio were selected to conduct the single factor experiments.Based on the results of single factor experiment,the extraction process was optimized by the Box-Behnken test design with the total flavonoids extraction rate as the response value.DPPH,ABTS and superoxide radical scavenging tests were used to compare the antioxidant capacity of the products obtained by ultrasonic method and reflux method.Results:The optimum condition was as follows:Rhamnella gilgitica Mansf.et Melch were extracted with 53.33%ethanol at the solid-liquid ratio of 1∶48.42 mL/g for 50.00 min at 54.66℃.Under these conditions,the extraction rate of total flavonoids was 8.16%.The results showed that the total flavonoids of Rhamnella gilgitica Mansf.et Melch had good antioxidant activity,and the antioxidation ability of ultrasonic extraction was better than that of reflux extraction.Conclusion:Ultrasonic extraction is a good way to extract the total flavonoids from Rhamnella gilgitica Mansf.et Melch and the extracted products have good antioxidant effect.
