Malat1通过激活p38 MAPK/p53信号通路调控缺氧/复氧AC16心肌细胞凋亡 被引量:6

Malat1 regulates hypoxia/reoxygenation-induced apoptosis by activating p38 MAPK/p53 signaling pathway in cardiomyocytes

摘要 目的:探讨长链非编码RNA肺癌代谢相关转录本1(Malat1)对缺氧/复氧(H/R)诱导的人AC16心肌细胞凋亡的影响及机制。方法:构建AC16心肌细胞H/R模型,采用pcDNA3.1-Malat1、si-Malat1质粒转染细胞的方式调控Malat1的表达水平。采用实时荧光定量PCR(qRT-PCR)技术检测细胞中Malat1、p38、p53、促凋亡基因Bax、Caspase-3及抑凋亡基因Bcl-2的表达,采用蛋白印迹法检测p38蛋白、磷酸化p38蛋白(p-p38)、p53蛋白、磷酸化p53蛋白(p-p53)的表达。结果:Malat1过表达增加了H/R处理所致心肌细胞内促凋亡相关基因Bax、Caspase 3及其蛋白的表达,而减少了抑凋亡相关基因Bcl-2及其蛋白的表达,同时p38、p-p38及p53、p-p53蛋白表达水平也显著上升。以上表达差异均具有统计学意义(P<0.05)。结论:Malat1可通过调控p38 MAPK/p53信号通路激活的方式参与调控H/R诱导的心肌细胞凋亡。 Objective:To investigate the effect of metastasis-associated lung adenocarcinoma transcript 1(Malat1)on apoptosis induced by hypoxia/reoxygenation(H/R)in human AC16 cardiomyocyte.Methods:The H/R model of AC16 cardiomyocyte was constructed,and Malat1-containing plasmids and si-Malat1 plasmids were used to regulate Malat1 expression.The mRNA expression levels of Malat1,p38,p53,and apoptosis-related genes(Bax,Bcl-2,Caspase-3)in cells were detected by real-time fluorescent quantitative PCR(qRT-PCR),and p-p38,p38,p-p53,p53 protein levels were de-tected by Western Blot.Results:Malat1 overexpression increased the mRNA and the protein expres-sion levels of pro-apoptotic related genes Bax and Caspase-3 in myocardial cells induced by H/R treat-ment,the mRNA and protein levels of anti-apoptotic related genes Bcl-2 were reduced,while p-p38/p38 and p-p53/p53 protein levels were also significantly increased.All differences were statistically significant(all P<0.05).Conclusion:Malat1 can participate in the regulation of H/R-induced apopto-sis by activating p38 MAPK/p53 signaling pathway in cardiomyocyte.
