摘要
秸秆是农林废弃物之一,高纤维素含量的秸秆是制造纤维素酶的潜在低成本原料。从木质纤维素生产纤维素酶的研究较成熟,但酶活性普遍很低,为了降低成本,提高纤维素酶活性,使用黑曲霉固态发酵秸秆生产纤维素酶。用碱预处理降低木质素含量,在底物中接种黑曲霉并孵育,考察了p H值,反应温度,底物浓度对酶活性的影响。研究结果显示,最高酶活性3.12U·m L-1,蛋白质含量0.34mg·m L-1,发酵时间为4d,水含量为75%。在酶表征中,最佳p H值为4,温度60℃。从酶促反应动力学所获得的60℃温度下的动力学参数为Vmax为5.82U·m L-1;Km为0.32mmol·L-1。
Straw is one of the agricultural and forestry wastes.Straw with high cellulose content is a potential low-cost raw material for cellulase production.The research on cellulase production from lignocellulose is mature,but the enzyme activity is generally low.In order to reduce the cost and improve the cellulase activity,Aspergillus niger is used to produce cellulase from solid-state fermentation straw.The effects of pH value,reaction temperature and substrate concentration on enzyme activity were investigated.The results showed that the highest enzyme activ-ity was 3.12 U·mL-1,protein content was 0.34 mg·mL-1,fermentation time was 4 days,and water content was 75%.In the enzyme characterization,the optimal pH is 4 and the temperature is 60℃.The kinetic parameters at 60℃obtained from the enzymatic reaction kinetics were Vmax5.82 u·mL-1 and km0.32 mmol·L-1.
